Clostridium sporogenes [CMCC (B) 64941]

Clostridium sporogenes CMCC(B)64941 is a strictly anaerobic, spore-forming Gram-positive bacillus obtained from the China Medical Culture Collection Center (CMCC). As a member of the Clostridium genus, this organism produces endospores that are highly resistant to heat and chemical disinfection, making it an important reference strain for validating sterilization processes and anaerobic culture methodologies.

The strain is cultured under strict anaerobic conditions using liquid thioglycollate medium (FT), Brucella broth, or CDC anaerobic blood agar. Incubation times range from 24 hours up to 7 days depending on the medium, as some strict anaerobes require extended incubation for full growth expression. On TSC egg yolk agar, this strain characteristically shows no blackening of colonies and no lecithinase precipitation rings, which is a key differential feature from pathogenic C. perfringens.

This CMCC reference strain is primarily intended for:

1. Preparation: Before activation, prepare suitable solid and liquid culture media and equipment. All operations must be conducted under biosafety protection and sterile conditions.
2. Resurrection: Remove the slant culture. Under sterile conditions, open the test tube by pushing the safety lock block downward along the arrow direction, gently press, and allow the lid to pop open. Remove the lid, then use a sterile inoculating loop to pick up an appropriate amount of bacterial lawn and inoculate into suitable solid or liquid culture media.
3. Incubation: Place cultures under appropriate anaerobic conditions:
   • Liquid FT medium: 30–35°C for 72 hours
   • Brucella broth: 36±1°C, anaerobic, 24–72 hours
   • CDC anaerobic blood agar: 36±1°C, anaerobic, 48–72 hours (up to 5–7 days for strict anaerobes)
4. Post-inoculation storage: Push the safety lock block upward to lock the lid, then store at appropriate temperature. Observe growth after cultivation.
5. Working stock maintenance: Store active strains at 2–8°C under anaerobic conditions. Pass regularly to prevent degeneration.

3. Follow appropriate biosafety guidelines for BSL-2 organisms. All operations must be conducted under biosafety protection and sterile conditions.
4. Anaerobic bacteria should be activated as soon as possible and then stored under anaerobic conditions at 2–8°C.
5. If contamination or significant changes in colony characteristics are detected, the strain shall not be used.
6. Strains beyond their expiration date must not be used; passage regularly to prevent degeneration or death.
