ELISA Intend Use
Mouse Cgref1 ELISA Kit is a Sandwich ELISA KIT detecting the concentration of Cell growth regulator with EF hand domain protein 1 in Mouse serum, plasma, tissue homogenates and other biological fluids.
PRINCIPLE OF THE ASSAY
This assay employs a two-site sandwich ELISA to quantitate CGREF1 in Mouse serum, plasma. An antibody specific for CGREF1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CGREF1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CGREF1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CGREF1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Detect Range
31.25 pg/mL - 2000 pg/mL. The standard curve concentrations used for the ELISA’s were 2000 pg/mL, 1000 pg/mL, 500 pg/mL, 250 pg/mL, 125 pg/mL, 62.5 pg/mL,
31.25 pg/mL, 0 pg/mL.
Sensitivity
The limit of detection of Mouse CGREF1 defined as the analyte concentration resulting in an absorbance significantly higher than that of the dilution medium (mean plus 2 standard deviations) was determined to be 15.6 pg/mL (mean of 6 independent assays).
Cgref1 Introduction
A full-length CGREF1 cDNA cloned from a human fetal brain cDNA library encodes a deduced 301-amino acid protein with a highly conserved putative Ca(2+)-binding EF-hand motif. The human and rat CGREF1 proteins share 65% overall sequence identity and nearly 100% in the EF-hand motif. The human protein has 4 clustered 17-amino acid repeats within the C terminus, suggesting a structure resembling 4 alpha-helical domains interrupted by distinct turns. 4 Northern blot analysis of rat tissues demonstrated expression of CGREF1 predominantly in whole brain and kidney, with limited expression in heart, lung, liver, and skeletal muscle and no expression in spleen and testis. Transfection assay experiments demonstrated that CGREF1 is able to inhibit growth in several cell lines.
